The researchers constructed genetically engineered mice that lack the ABHD6 enzyme in a localized region of the brain, a specific group of hypothalamic neurons. The first author of the paper, Alexandre Fisette, explains, “These mice are exactly the same as normal mice under normal accommodation and food conditions. However, when challenged, they are unable to adapt. Dr. Thierry Alquier, a co-author of the paper and a professor at the University of Montreal Hospital Research Center, commented, “It is a mechanism we have never suspected. Stephanie Fulton, a co-author of the paper and a researcher at the University of Montreal Hospital Research Center, stressed, "We now know that ABHD6 plays a completely different role in certain neurons in the hypothalamus. Blocking this enzyme at this position promotes obesity. Blocking it elsewhere in the body can have beneficial effects."

A variety of signaling and neural networks are involved in regulating energy balance and maintaining a stable body weight. "We have shown that the ABHD6 enzyme plays a key role in maintaining homeostasis in specific neurons in the hypothalamus," the researchers explained.
