How to quickly detect the accuracy of the pipette?

Pipettes are one of the most commonly used instruments in the laboratory and one of the instruments most likely to affect the results of the experiment, ensuring its accuracy is essential. So what can we do if we suspect the accuracy of the pipette during the pipette? Since it is necessary to ensure the efficiency and time of the experiment and other related issues, it is impossible for us to go to the manufacturer or dealer to help detect the problem every time the pipette has a problem. Therefore, we only have to detect the pipette by ourselves. For details, please refer to the following methods:

First, we need to determine if there is a leak in the pipette.
1. Detection method:
1) Visual inspection: first pipette the liquid using the pipette to be tested, then let it stand vertically for 15 seconds to see if there is a slow flow of droplets. If there is a droplet flowing slowly, it means that the pipette has air leakage.
2) Pressure pump detection: Use a dedicated pressure pump to detect the pressure and determine if the pipette is leaking.

2. If there is a leak in the pipette, then we need to analyze the possible causes of the leak:
First, check if the tip matches? It is recommended to use original matching tips. If it is not the original matching tip, it is best to consult the manufacturer or agent to see if the tip you are using matches the pipette used.
Secondly, is there any tightening when assembling the tip? It is very likely that the pipette leaks due to the lack of tightening when the tip is loaded.
Finally, if no problem is found in either of the above cases, it is likely that the internal airtightness of the pipette is not good. At this time, you need to contact the pipette company maintenance engineer.

When we exclude the first case above, we are going to check the second case:

We need to check if the temperature of the pipette is different from the temperature difference between the pipette and the tip, which affects the accuracy of the pipette.
In general, when the temperature of the removed liquid does not match the temperature of the tip and the pipette, the following two differences occur:

1. When the liquid temperature is higher than the tip, the volume of the removed liquid will be too large;

2. When the liquid temperature is lower than the tip, the volume of the liquid removed will be too small.

If the second case is still excluded, then we have to check the third case below:

Is there a very significant difference between the density of the liquid sample and the water?
In general, the pipette calibration before leaving the factory is tested with water as a standard solution, so if the specific gravity of the liquid to be treated is very different, the accuracy and accuracy given may not be met. An error has occurred.

After eliminating all the above problems, we should also pay attention to the liquid absorption speed is not too fast, the liquid absorption speed will produce backlash and air bubbles too fast, resulting in inaccurate pipetting volume, and ultimately affect the accuracy of the pipette.
