Aggravating factors for Alzheimer's disease were found

US researchers published a report in the new British journal Nature on the 25th that meningeal lymphatic dysfunction may be a key factor leading to the aggravation of Alzheimer's disease and age-related cognitive dysfunction. This discovery is expected to provide new ideas for the treatment of related diseases.

The academic community once thought that the brain lacks the lymphatic system. But in 2014, American researchers discovered the presence of meningeal lymphatics. In the new study, researchers at the University of Virginia School of Medicine who had discovered meningeal lymphatics and researchers at Virginia Tech confirmed that meningeal lymphatics can drain macromolecules from the cerebrospinal fluid and interstitial fluids of the central nervous system to the cervical lymph nodes.

Studies have found that the damage of meningeal lymphatic function in adult mice leads to a decline in learning and memory; some age-related cognitive dysfunction in aged mice is associated with severe damage to meningeal lymphatic function. In elderly mice, after receiving vascular endothelial growth factor therapy, the ability of meningeal lymphatic vessels to clear macromolecules in cerebrospinal fluid and interstitial fluid is enhanced, and the learning and memory ability is also improved.

Jennifer Munson, an assistant professor of biomedical engineering at Virginia Tech University in the United States, said that during aging, fluid flow in the brain slows down, sometimes half as fast as when young. The meningeal lymphatics discharge the proteins that cause Alzheimer's disease along with other cellular waste, and a slower flow rate causes this protein to accumulate. Existing studies suggest that Alzheimer's disease is associated with excessive accumulation of beta-amyloid in the brain.

Researchers have developed a hydrogel that contains molecules that dilate the meningeal lymphatics that can spread through the brain and into the meninges. Researchers believe that similar hydrogels in the future are expected to be non-invasive drugs for the treatment of Alzheimer's disease and age-related cognitive dysfunction.
